This gene encodes one of the three enolase isoenzymes found in mammals. This isoenzyme, a homodimer, is found in mature neurons and cells of neuronal origin. A switch from alpha enolase to gamma enolase occurs in neural tissue during development in rats and primates. Sample Type: Serum, plasma, tissue homogenates, Intended Use: For the quantitative determination of human neuron-specific enolase (NSE) concentrations in serum, plasma, tissue homogenates. Sensitivity: 0.195ng/ml, Range: 0.78-50ng/ml, Specificity: This assay has high sensitivity and excellent specificity for detection of human NSE. No significant cross-reactivity or interference between human NSE and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for NSE has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NSE present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NSE is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NSE bound in the initial step. The color development is stopped and the intensity of the color is measured. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
