Mouse CD6 is a type I transmembrane glycoprotein belonging to the group B scavenger receptor cysteine-rich (SRCR) superfamily. This 626-amino-acid murine protein consists of a 24-amino-acid signal sequence, a 372-amino-acid extracellular region with three conserved SRCR domains, and a 204-amino-acid cytoplasmic tail lacking intrinsic catalytic activity. Mouse CD6 shares 70% full-length amino acid identity with its 668-amino-acid human counterpart. It is lowly expressed on immature thymocytes and highly expressed on mature thymocytes, as well as in most peripheral T cells, partial B cells and neuronal cells. CD6 serves as a dual functional adhesion receptor and T cell costimulatory molecule. Its domain 3 binds ALCAM/CD166 to stabilize immunological synapses and modulate thymocyte development, while domain 1 interacts with CD318. It potentiates T cell activation and proliferation via regulating TCR/CD3 signaling. Upon T cell activation, CD6 undergoes multi-site phosphorylation to initiate intracellular signaling, though its downstream pathways remain incompletely defined. It also binds bacterial LPS and LTA to modulate inflammation and acts as a key target for autoimmune and allograft rejection therapies. Protein function: Cell adhesion molecule that mediates cell-cell contacts and regulates T-cell responses via its interaction with ALCAM/CD166. Contributes to signaling cascades triggered by activation of the TCR/CD3 complex (PubMed:24584089). Functions as a costimulatory molecule, promotes T-cell activation and proliferation. Contributes to the formation and maturation of the immunological synapse. Functions as a calcium-dependent pattern receptor that binds and aggregates both Gram-positive and Gram-negative bacteria. Binds both lipopolysaccharide (LPS) from Gram-negative bacteria and lipoteichoic acid from Gram-positive bacteria. LPS binding leads to the activation of signaling cascades and down-stream MAP kinases. Mediates activation of the inflammatory response and the secretion of pro-inflammatory cytokines in response to LPS [The UniProt Consortium]
