NOV (CCN3) is a secreted matricellular protein belonging to the conserved CCN family, featuring four typical modular domains: IGFBP, vWF type C, thrombospondin-1, and a C-terminal cysteine knot domain. Human CCN3 shares 80% amino acid identity with its mouse homolog, indicating high cross-species conservation. Widely expressed during development, CCN3 is abundant in muscle, endothelial cells, nervous tissues, adrenal cortex and chondrocytes. Full-length CCN3 is secreted into the extracellular matrix, binding integrin alphavbeta3 and alpha5beta1 to mediate endothelial adhesion, chemotaxis and angiogenesis, and suppresses cell proliferation. It modulates hematopoietic, osteogenic and chondrogenic cell self-renewal and maturation via Notch and BMP signaling, and interacts with multiple partners including Notch1, CCN2, fibulin 1C, Connexin43 and S100A4. Truncated nuclear CCN3 functions as a transcriptional repressor to promote cell proliferation. It also inhibits myogenic gene expression and is closely associated with the progression of Wilm's tumor and nephroblastoma. In endothelial cells, supports cell adhesion, induces directed cell migration (chemotaxis) and promotes cell survival. Also plays a role in cutaneous wound healing acting as integrin receptor ligand.
