Product information "Human sC5b-9 (Soluble Terminal Complement Complex C5b-9) Microsample ELISA Kit"
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human sC5b-9. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human sC5b-9. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human sC5b-9, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human sC5b-9 in the samples is then determined by comparing the OD of the samples to the standard curve.Detection Range: 78.13-5000 ng/mL. Sensitivity: 19.9 ng/mL. Assay Type: Sandwich. Standard: 5000 ng/mL. Sample Type: sserum, plasma and other biological fluids. Assay duration: 3.5h. Protein Function: Pore-forming component of the membrane attack complex (MAC), a multiprotein complex activated by the complement cascade, which inserts into a target cell membrane and forms a pore, leading to target cell membrane rupture and cell lysis (PubMed:22832194, PubMed:26841837, PubMed:26841934, PubMed:27052168, PubMed:30552328, PubMed:6177822, PubMed:9212048, PubMed:9634479). The MAC is initiated by proteolytic cleavage of C5 into complement C5b in response to the classical, alternative, lectin and GZMK complement pathways (PubMed:9212048, PubMed:9634479). The complement pathways consist in a cascade of proteins that leads to phagocytosis and breakdown of pathogens and signaling that strengthens the adaptive immune system (PubMed:9212048, PubMed:9634479). Constitutes the pore-forming subunit of the MAC complex: during MAC assembly, C9 associates with the C5b8 intermediate complex, and polymerizes to complete the pore (PubMed:26841934, PubMed:30111885, PubMed:30552328, PubMed:34752492, PubMed:4055801, PubMed:6177822) [The Uniprot Consortium]
