The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human IP10. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human IP10. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human IP10, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human IP10 in the samples is then determined by comparing the OD of the samples to the standard curve.Detection Range: 15.63-1000 pg/mL. Sensitivity: 6 pg/mL. Assay Type: Sandwich. Standard: 1000 pg/mL. Sample Type: sserum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Assay duration: 3.5h. Protein Function: Component of the membrane attack complex (MAC), a multiprotein complex activated by the complement cascade, which inserts into a target cell membrane and forms a pore, leading to target cell membrane rupture and cell lysis (PubMed:22832194, PubMed:26841837, PubMed:27052168, PubMed:30552328, PubMed:3335508). The MAC is initiated by proteolytic cleavage of C5 into complement C5b in response to the classical, alternative, lectin and GZMK complement pathways (PubMed:22832194, PubMed:30552328, PubMed:3335508). The complement pathways consist in a cascade of proteins that leads to phagocytosis and breakdown of pathogens and signaling that strengthens the adaptive immune system (PubMed:22832194, PubMed:30552328, PubMed:3335508). C7 serves as a membrane anchor (PubMed:30552328). During MAC assembly, associates with C5b and C6 to form the C5b-7 complex, a key lipophilic precursor of the MAC complex, which associates with the outer leaflet and reduces the energy for membrane bending (PubMed:30552328, PubMed:32569291) [The Uniprot Consortium]
