The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human BMPR2. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human BMPR2. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human BMPR2, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human BMPR2 in the samples is then determined by comparing the OD of the samples to the standard curve.Detection Range: 0.32-20 ng/mL. Sensitivity: 0.123 ng/mL. Assay Type: Sandwich. Standard: 20 ng/mL. Sample Type: sTissue homogenates, cell lysates and other biological fluids.. Assay duration: 3.5h. Protein Function: On ligand binding, forms a receptor complex consisting of two type II and two type I transmembrane serine/threonine kinases. Type II receptors phosphorylate and activate type I receptors which autophosphorylate, then bind and activate SMAD transcriptional regulators. Can also mediate signaling through the activation of the p38MAPK cascade (PubMed:12045205). Binds to BMP7, BMP2 and, less efficiently, BMP4. Binding is weak but enhanced by the presence of type I receptors for BMPs. Mediates induction of adipogenesis by GDF6. Promotes signaling also by binding to activin A/INHBA (PubMed:24018044) [The Uniprot Consortium]
