Granzyme B (Human) Chemiluminescent ELISA Detection Kit

The Granzyme B (Human) Chemiluminescent ELISA Detection Kit is a sandwich ELISA designed for detecting and quantifying human Granzyme B in cell culture medium. This kit comes with enough capture and detection antibodies, Granzyme B standard, and detection reagents for 100 or 5 x 100 reactions. Figure 1. Granzyme B (Human) Chemiluminescent ELISA Detection Kit schematic. A 96-well plate is coated with an anti-Granzyme B capture antibody. After coating and blocking, samples containing Granzyme B are added. Next, unbound Granzyme B is washed away, and the plate is incubated with the biotinylated Granzyme B detection antibody. After another wash, streptavidin-HRP is added. After a final wash, ELISA ECL substrate is added to produce chemiluminescence that can be measured using a chemiluminescence reader. The chemiluminescence signal is proportional to the amount of Granzyme B in the samples. Granzyme B (GrB) is a serine protease secreted by immune cells, like CTLs (cytotoxic T lymphocytes) and NK (natural killer) cells, to kill tumor and virus-infected cells. Immune cells can target and kill target cells through several mechanisms, one of them being the release of cytotoxic granules. These cytotoxic granules contain several proteases, such as granzyme B. Granzyme B acts on pro-caspase 3 and 7, triggering caspase dependent cell death. The amount of granzyme B present in co-culture is a typical measure to determine the cytotoxic potential of CAR (chimeric antigen receptor)-T and NK cells. The availability of simple quantitative assays is crucial during the functional validation of cell therapies.
