Elafin (PI3) BioAssay(TM) ELISA Kit (Human)

This gene encodes an elastase-specific inhibitor that functions as an antimicrobial peptide against Gram-positive and Gram-negative bacteria. The protein contains a WAP-type four-disulfide core (WFDC) domain, and is thus a member of the WFDC domain family. Most WFDC gene members are localized to chromosome 20q12-q13 in two clusters: centromeric and telomeric. This gene belongs to the centromeric cluster. Expression of this gene is upgulated by bacterial lipopolysaccharides and cytokines. Sample Type: Serum, plasma and tissue homogenates, Intended Use: For the quantitative determination of human elafin (PI3) concentrations in serum, plasma and tissue homogenates. Sensitivity: 0.156ng/ml, Range: 0.625-40ng/ml, Specificity: This assay has high sensitivity and excellent specificity for detection of human PI3. No significant cross-reactivity or interference between human PI3 and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PI3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PI3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PI3 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PI3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
