This gene is one of several CC cytokine genes clustered on the q-arm of chromosome 17. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene functions as a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. This cytokine is one of the major HIV-suppressive factors produced by CD8+ cells. It functions as one of the natural ligands for the chemokine receptor CCR5 and it suppresses in vitro replication of the R5 strains of HIV-1, which use CCR5 as a coreceptor. Sample Type: Serum, plasma and tissue homogenates, Intended Use: For the quantitative determination of guinea pig C-C motif chemokine 5 (CCL5) concentrations in serum, plasma and tissue homogenates. Sensitivity: 0.081ng/ml, Range: 0.156-10ng/ml, Specificity: This assay has high sensitivity and excellent specificity for detection of guinea pig CCL5. No significant cross-reactivity or interference between guinea pig CCL5 and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CCL5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CCL5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CCL5 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CCL5 bound in the initial step. The color development is stopped and the intensity of the color is measured. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
