The ASAP1 (Arf-GAP with SH3 Domain, ANK Repeat and PH Domain-Containing Protein 1) BioAssay(TM) ELISA Kit (Human) is a quantitative sandwich assay for the detection of ASAP1 in human serum, plasma and other biological fluids. Cell lysates and tissue homogenates, though not tested, may potentially be used as samples. No significant cross-reactivity or interference between ASAP1 and analogues was observed. Anti-ASAP1 antibody was pre-coated onto 96-well plates. The biotin conjugated anti-ASAP1 antibody, was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were, used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the ASAP1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of ASAP1 can be calculated. For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap.
