This gene is a member of the angiopoietin/angiopoietin-like gene family and encodes a glycosylated, secreted protein with a fibrinogen C-terminal domain. This gene is induced under hypoxic conditions in endothelial cells and is the target of peroxisome proliferation activators. The encoded protein is a serum hormone directly involved in regulating glucose homeostasis, lipid metabolism, and insulin sensitivity and also acts as an apoptosis survival factor for vascular endothelial cells. The encoded protein may play a role in several cancers and it also has been shown to prevent the metastatic process by inhibiting vascular activity as well as tumor cell motility and invasiveness. Decreased expression of this protein has been associated with type 2 diabetes. Alternatively spliced transcript variants encoding different isoforms have been described. This gene was previously referred to as ANGPTL2 but has been renamed ANGPTL4. Sample Type: Serum, plasma, tissue homogenates, Intended Use: For the quantitative determination of human angiopoietin-related protein 4 (ANGPTL4) concentrations in serum, plasma, tissue homogenates. Sensitivity: 0.39ng/ml, Range: 1.56-100ng/ml, Specificity: This assay has high sensitivity and excellent specificity for detection of human ANGPTL4. No significant cross-reactivity or interference between human ANGPTL4 and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ANGPTL4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ANGPTL4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ANGPTL4 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ANGPTL4 bound in the initial step. The color development is stopped and the intensity of the color is measured. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
