This gene encodes a complex glycoprotein secreted in plasma. The precursor is proteolytically processed into distinct functioning proteins: alpha-1-microglobulin, which belongs to the superfamily of lipocalin transport proteins and may play a role in the regulation of inflammatory processes, and bikunin, which is a urinary trypsin inhibitor belonging to the superfamily of Kunitz-type protease inhibitors and plays an important role in many physiological and pathological processes. This gene is located on chromosome 9 in a cluster of lipocalin genes. Sample Type: Serum, plasma, urine, tissue homogenates, Intended Use: For the quantitative determination of human alpha-1-microglobulin/bikunin precursor (AMBP) concentrations in serum, plasma, urine, tissue homogenates. Sensitivity: 0.039ng/ml, Range: 0.156-10ng/ml, Specificity: This assay has high sensitivity and excellent specificity for detection of human AMBP. No significant cross-reactivity or interference between human AMBP and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for AMBP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any AMBP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for AMBP is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AMBP bound in the initial step. The color development is stopped and the intensity of the color is measured.
