Product information "TRIM24 (TIF1) TR-FRET Assay Kit"
The TRIM24 (TIF1) TR-FRET Assay Kit is designed to measure the binding of TRIM24 (tripartite motif-containing 24), also known as TIF1, to its substrate for screening and profiling applications using TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer). It utilizes a Terbium-labeled donor and a dye-labeled acceptor to complete the TR-FRET pairing. The TRIM24 TR-FRET Assay Kit comes in a convenient 384-well format, with enough purified TRIM24 (896-1014 amino acids), Ligand, Tb-Labeled Donor and Dye-Labeled Acceptor, Bromodomain Ligand 3 and assay buffer for 400 reactions. The assay also includes Non-Acetylated Ligand 3 as control for the specificity of binding. Figure 1: Illustration of the assay principle. A sample containing terbium-labeled donor, dye-labeled acceptor, TRIM24, substrate, and an inhibitor is incubated. When TRIM24 binds to its substrate, energy transfer occurs due to the proximity of the donor and acceptor. Disruption of the binding results in decrease of energy transfer. TRIM24 (tripartite motif containing 24), also known as TIF1 (transcriptional intermediary factor 1 alpha), is a protein involved in several processes such as transcriptional control, chromatin regulation, and stress responses. It has E3 ubiquitin ligase activity and can also work as a transcriptional coactivator. High protein levels have been linked to cancer, such as gastric cancer (GC). Studies have identified that in GC, TRIM24 binds to NRBP1 (nuclear receptor binding protein 1), leading to its ubiquitination and degradation. The use of TRIM24 inhibitors can potentially benefit cancer patients.
