Product information "KLK11 Fluorogenic Assay Kit"
The KLK11 Fluorogenic Assay Kit is designed to measure KLK11 (Kallikrein-11) protease activity for screening and profiling applications. The assay kit comes in a convenient 96-well format, with enough recombinant (amino acids 54-282), fluorogenic AMC-based peptide substrate, and assay buffer for 100 enzyme reactions. Figure 1: Illustration of the assay principle. A peptide substrate conjugated to AMC is incubated with KLK11. The conjugated AMC displays only weak fluorescence. Upon proteolysis, AMC is no longer quenched and exhibits strong blue fluorescence. The increase in fluorescence is proportional to the KLK11 activity. KLK11 (kallikrein 11), also know as TLSP (Trypsin-like serine protease), is a secreted serine endopeptidase that participates in skin desquamation. Abnormal expression of this protein can result in skin barrier-related disorders. It can be found in multiple tissues, such as the brain, skin, breast, prostate and intestine. It can be found at high levels in the serum of breast and prostate cancer patients, and it is considered a favorable prognostic factor. However, in other cancer types it seems to correlate with a poor prognosis. In a model of colorectal cancer (CRC) the knockdown of KLK11 lead to inhibition of cell growth and increased in apoptosis. A deeper understanding of the role in health and disease of this protein may open new therapeutic opportunities in oncology.
