Fibroblast Activation Protein, or FAP, is a 170kD dimer with dipeptidase activity. FAP expression is seen on activated stromal fibroblasts in >90% of all human carcinomas, suggesting a role in the development, growth and metastasis of carcinomas. It is also involved in the control of wound healing and tissue repair. The FAP Fluorescent Activity Assay kit is designed to measure FAP activity using purified FAP or cell extracts containing FAP. It comes in a convenient 96-well format, with purified FAP enzyme, FAP substrate, and FAP assay buffer for 100 enzyme reactions. The key to the FAP Fluorescent Activity Assay is the fluorogenic substrate. Using this kit, only one simple step on a microtiter plate is required for FAP reactions. The FAP fluorometric substrate is incubated with a sample containing FAP enzyme to produce a fluorophore that can then be measured using a fluorescence reader. Kit Components: F4208-50A1: Microtiter Plate, 1x96wells, F4208-50A2: DPP assay buffer, 1x10ml , F4208-50A3: Fluorogenic DPP substrate (0.5mM), 1x100ul Supplied as a liquid in DMSO., F4208-50A4: FAP Recombinant Human, 1x30ug, F4208-50A5: Fluorescent AMC Standard (50uM), 1x500ul , APPLICATIONS: Suitable for studying enzyme kinetics and screening small molecular inhibitors for drug discovery and HTS applications. Other applications not tested. Storage and Stability: Store F4208-50A1 at RT. Store F4208-50A2 and F4208-50A5 at -20°C. Store F4208-50A3 and F4208-50A4 at -70°C. Stable for 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
