The Enzolution(R) CD38 Assay System is intended for use with the Transcreener(R) ADPR FP Assay Kit (Part #3031) to measure enzymatic activity for CD38 (Also known as Cluster of Differentiation 38). CD38 is a multifunctional, membrane-bound enzyme that modulates beta-Nicotinamide adenine dinucleotide (NAD) levels by degrading it to adenosine diphosphate ribose (ADPR) and nicotinamide. The Transcreener ADPR FP assay uses a coupling enzyme to convert ADPR to AMP, which is then detected using a far-red, competitive fluorescence polarization (FP) assay. It is in a single addition, endpoint mix-and-read format in which enzyme reactions are quenched by addition of the detection reagents. The assay has been optimized and extensively validated for high throughput screening (HTS) and inhibitor dose response measurements using most multimode plate readers. Protein Function: Synthesizes cyclic ADP-ribose (cADPR), a second messenger for glucose-induced insulin secretion (PubMed:7961800, PubMed:8253715). Synthesizes the Ca(2+) mobilizer nicotinate-adenine dinucleotide phosphate, NAADP(+), from 2'-phospho-cADPR and nicotinic acid, as well as from NADP(+) and nicotinic acid. At both pH 5.0 and pH 7.4 preferentially transforms 2'-phospho-cADPR into NAADP(+), while preferentially cleaving NADP(+) to cADPR and ADPRP rather than into NADDP(+) (PubMed:16690024).
