Product Description: Caspase 10 (CASP10) is a member of the cysteine-aspartic acid protease (caspase) family, playing a crucial role in the execution phase of cell apoptosis. It is encoded by the CASP10 gene and exists as an inactive proenzyme that undergoes proteolytic processing to produce two subunits that dimerize to form the active enzyme. The protein itself is processed by caspase 8 and cleaves and activates caspases 3 and 7, highlighting its central role in the apoptosis pathway. The protein contains two death effector domains (DED) and is involved in the activation cascade of caspases responsible for apoptosis execution, interacting with FADD, CFLAR, Caspase 8, and other proteins. Mutations in the CASP10 gene are associated with apoptosis defects seen in type II autoimmune lymphoproliferative syndrome, a condition characterized by an overactive immune response and lymphoproliferation. Protein Function: Involved in the activation cascade of caspases responsible for apoptosis execution. Recruited to both Fas- and TNFR-1 receptors in a FADD dependent manner. May participate in the granzyme B apoptotic pathways. Cleaves and activates effector caspases CASP3, CASP4, CASP6, CASP7, CASP8 and CASP9. Hydrolyzes the small- molecule substrates, Tyr-Val-Ala-Asp-, -AMC and Asp-Glu-Val-Asp-, -AMC , Isoform 7 can enhance NF-kappaB activity but promotes only slight apoptosis , Isoform C is proteolytically inactive [The Uniprot Consortium]
