Product Description: Tubulin tyrosine ligase-like protein 12 (TTLL12) is a 644-amino acid enzyme belonging to the tubulin tyrosine ligase-like (TTLL) family, primarily involved in regulating post-translational modifications of tubulin, such as detyrosination and polyglutamylation, as well as histone modifications, notably promoting histone H4 trimethylation at lysine 20 (H4K20me3). Structurally, TTLL12 contains a SET-like domain, linked to histone methylation, and a TTL-like domain, associated with tubulin modifications, and is localized in the cytosol and plasma membrane. Functionally, it plays a crucial role in mitotic regulation, chromosome stability, and negatively regulates type I interferon-mediated signaling, particularly during RNA virus infection by modulating the DDX58/RIG-I pathway. Clinically, elevated TTLL12 expression has been correlated with poor prognosis in ovarian cancer, where high levels are associated with advanced disease stage and reduced overall and disease-free survival, suggesting its potential as a prognostic biomarker. Protein Function: Negatively regulates post-translational modifications of tubulin, including detyrosination of the C-terminus and polyglutamylation of glutamate residues (PubMed:20162578, PubMed:23251473). Also, indirectly promotes histone H4 trimethylation at 'Lys-20' (H4K20me3) (PubMed:23251473). Probably by controlling tubulin and/or histone H4 post-translational modifications, plays a role in mitosis and in maintaining chromosome number stability (PubMed:20162578, PubMed:23251473). During RNA virus-mediated infection, acts as a negative regulator of the RIG-I pathway by preventing MAVS binding to TBK1 and IKBKE (PubMed:28011935) [The Uniprot Consortium]
