It serves as a ligand for osteoprotegerin and plays a crucial role in osteoclast differentiation and activation. TNFSF11 acts as a dendritic cell survival factor and regulates T-cell-dependent immune responses, with T cell activation inducing its expression. The protein enables identical protein binding activity, receptor ligand activity, and tumor necrosis factor receptor superfamily binding. TNFSF11 activates the antiapoptotic kinase AKT/PKB through a signaling complex involving SRC kinase and TRAF6, suggesting a role in cell apoptosis regulation. Regarding disease associations, disruption of TNFSF11 in mice results in severe osteopetrosis and absence of osteoclasts, along with defects in early T and B lymphocyte differentiation.
