Product Description: Transcription factor 20 (TCF20) is a nuclear protein encoded by the TCF20 gene that functions primarily as a transcriptional coactivator, binding to regulatory DNA elements such as the platelet-derived growth factor-responsive element in the matrix metalloproteinase 3 (MMP3) promoter and controlling stromelysin expression. TCF20 displays DNA-binding and transactivation activities, enhancing the function of multiple transcription factors including JUN, SP1, PAX6, and others, thereby influencing a wide range of physiological processes such as DNA damage repair, epigenetic gene regulation, protein modification, signal transduction, cellular proliferation, differentiation, apoptosis, neuronal and synaptic development, immune response, and lipid metabolism. The protein contains several functional domains, including nuclear localization sequences, AT-hook, DNA-binding, PHD, and zinc-finger domains, and alternative splicing results in different isoforms. Pathogenic variants in TCF20, particularly those causing loss of key domains, are associated with neurodevelopmental disorders, as they disrupt its regulatory functions and affect the activity of related transcription factors, leading to developmental and neurological abnormalities.
