Product Description: Transaldolase 1 (TALDO1) is a cytosolic enzyme crucial for the non-oxidative phase of the pentose phosphate pathway, where it catalyzes the reversible transfer of a three-carbon dihydroxyacetone unit from sedoheptulose-7-phosphate to glyceraldehyde-3-phosphate, producing erythrose-4-phosphate and fructose-6-phosphate. Structurally, human transaldolase consists of 337 amino acids forming a single alpha/beta barrel domain, with the active site containing lysine-142, glutamate-106, and aspartate-27, which are essential for its catalytic mechanism involving Schiff base formation and proton transfer. The enzyme functions as a dimer and is vital for generating ribose-5-phosphate for nucleic acid synthesis and NADPH for lipid biosynthesis. Deficiency or mutations in TALDO1 can disrupt the pentose phosphate pathway, leading to transaldolase deficiency, a rare metabolic disorder characterized by liver dysfunction, coagulopathy, and multi-organ involvement due to impaired redox balance and nucleotide synthesis. Protein Function: Catalyzes the rate-limiting step of the non-oxidative phase in the pentose phosphate pathway. Catalyzes the reversible conversion of sedheptulose-7-phosphate and D-glyceraldehyde 3-phosphate into erythrose-4-phosphate and beta-D-fructose 6-phosphate (PubMed:18687684, PubMed:8955144).
