STXBP3 interacts specifically with syntaxins to control SNARE complex assembly, essential for processes like insulin-stimulated GLUT4 vesicle fusion in adipocytes and muscle cells, thereby regulating glucose uptake. Structurally, STXBP3 modulates syntaxin conformation, influencing vesicle trafficking and exocytosis efficiency. It also plays roles in neutrophil degranulation and platelet aggregation by negatively regulating calcium-dependent exocytosis. Importantly, STXBP3-mediated regulation of vesicle fusion impacts insulin secretion and glucose homeostasis, linking it to metabolic disorders such as type 2 diabetes. Additionally, its interaction with ubiquitin and involvement in endosomal trafficking suggest broader roles in membrane protein sorting. Dysregulation of STXBP3 function can impair insulin secretion and vesicle trafficking, contributing to metabolic disease pathogenesis.
