Product Description: Syntaxin 16 (STX16) is a Qa-SNARE protein primarily localized to the trans-Golgi network (TGN) and late endosomes, where it mediates vesicular transport and membrane fusion events essential for intracellular trafficking. It contains a conserved domain structure typical of syntaxins, including an N-terminal peptide motif that binds the SM protein mVps45, facilitating SNARE complex formation. STX16 plays critical roles in recycling cargo such as the cystic fibrosis transmembrane conductance regulator (CFTR) and maintaining epithelial cell junctions by regulating E-cadherin trafficking. Additionally, STX16 is involved in autophagy by promoting transport of ATG9a-containing vesicles to autophagosomes and facilitating autolysosome formation. Functionally, STX16 is essential for epithelial morphogenesis, influencing lumen formation and transepithelial resistance. Dysregulation of STX16 impairs cell-cell junction integrity, leading to defects in epithelial barrier function and abnormal tissue organization, which may contribute to diseases involving epithelial dysfunction and impaired autophagy.
