Product Description: Sperm flagellar 2 (SPEF2), encoded by the SPEF2 gene, is a 128 kDa protein comprising 1135 amino acids, critical for ciliogenesis and sperm flagellum assembly. It contains an N-terminal domain, coiled-coil regions, and a dynein heavy chain domain, facilitating its role in microtubule organization and dynein arm complex formation within motile cilia and sperm flagella. SPEF2 localizes to the sperm flagellum and ciliary axonemes, where it regulates microtubule sliding and motility. It interacts with components like IFT20 and DNAH1, essential for intraflagellar transport and structural integrity. SPEF2 is predominantly expressed in testis and ciliated tissues, underscoring its reproductive and respiratory functions. Mutations in SPEF2 are linked to primary ciliary dyskinesia 43 (CILD43), characterized by chronic respiratory infections, situs inversus, and male infertility due to impaired sperm motility (spermatogenic failure 20, SPGF20). Protein Function: Required for correct axoneme development in spermatozoa. Important for normal development of the manchette and sperm head morphology. Essential for male fertility. Plays a role in localization of the intraflagellar transport protein IFT20 to the manchette, suggesting function as an adapter for dynein-mediated protein transport during spermatogenesis (PubMed:31048344, PubMed:31151990, PubMed:31278745). Also plays a role in bone growth where it seems to be required for normal osteoblast differentiation (By similarity) [The Uniprot Consortium]
