Product Description: Glutaminyl-peptide cyclotransferase (QPCT) is an enzyme that catalyzes the intramolecular cyclization of N-terminal glutamine and glutamic acid residues into pyroglutamate residues (pGlu), a posttranslational modification known as pyroglutamylation. This process is crucial for the maturation and stability of various hormones and secretory proteins, such as gonadotropin-releasing hormone (GnRH), thyrotropin-releasing hormone (TRH), and neurotensin. QPCT is encoded by the QPCT gene and is highly expressed in the pituitary, hypothalamus, and other parts of the brain. The enzyme plays a significant role in diseases such as Alzheimer's disease, where it contributes to the formation of pyroglutamate-modified amyloid peptides, and in cancer, where it is involved in the CD47-SIRPalpha pathway, affecting immune surveillance. Protein Function: Responsible for the biosynthesis of pyroglutamyl peptides. Has a bias against acidic and tryptophan residues adjacent to the N-terminal glutaminyl residue and a lack of importance of chain length after the second residue. Also catalyzes N-terminal pyroglutamate formation. In vitro, catalyzes pyroglutamate formation of N-terminally truncated form of APP amyloid-beta peptides [Glu-3]-amyloid-beta. May be involved in the N-terminal pyroglutamate formation of several amyloid-related plaque-forming peptides [The Uniprot Consortium]
