Product Description: Proteinase 3 (PRTN3) is a serine protease enzyme primarily expressed in neutrophil granulocytes. It plays a role in the proteolytic generation of antimicrobial peptides and is the target of anti-neutrophil cytoplasmic antibodies (ANCAs) of the c-ANCA class, commonly found in granulomatosis with polyangiitis (GPA). PRTN3 is externalized during apoptosis and interacts with phospholipid scramblase 1 (PLSCR1), which is involved in the bidirectional movement of plasma membrane phospholipids. This externalization can interfere with macrophage clearance of apoptotic neutrophils, contributing to the pathogenesis of ANCA-associated vasculitis and other inflammatory diseases. High levels of membrane PR3 expression are associated with chronic inflammatory diseases such as vasculitides and rheumatoid arthritis. Protein Function: Serine protease that degrades elastin, fibronectin, laminin, vitronectin, and collagen types I, III, and IV (in vitro) (PubMed:2033050, PubMed:28240246, PubMed:3198760). By cleaving and activating receptor F2RL1/PAR-2, enhances endothelial cell barrier function and thus vascular integrity during neutrophil transendothelial migration (PubMed:23202369). Plays a role in neutrophil transendothelial migration, probably when associated with CD177 (PubMed:22266279).
