Product Description: Polymeric immunoglobulin receptor (PIGR) is a transmembrane glycoprotein critical for mucosal immunity. Encoded by the PIGR gene, it facilitates the transcytosis of polymeric immunoglobulins A (IgA) and M (IgM) across epithelial cells. PIGR consists of five extracellular immunoglobulin-like domains, a transmembrane region, and a cytoplasmic tail. It binds to dimeric IgA or pentameric IgM at the basolateral surface of epithelial cells, internalizes the complex, and transports it to the apical membrane. Upon release, the extracellular domain of PIGR (called the secretory component) remains attached to IgA/IgM, forming secretory immunoglobulins (sIgA/sIgM) that protect mucosal surfaces from pathogens. Dysregulation of PIGR is linked to diseases such as inflammatory bowel disease (IBD) and chronic rhinosinusitis, where reduced expression compromises mucosal defense. Additionally, aberrant PIGR expression is observed in cancers, including colorectal and hepatocellular carcinomas, correlating with tumor progression and immune evasion. Genetic variants in PIGR may also predispose individuals to autoimmune disorders like systemic lupus erythematosus. Protein Function: Mediates selective transcytosis of polymeric IgA and IgM across mucosal epithelial cells. Binds polymeric IgA and IgM at the basolateral surface of epithelial cells. The complex is then transported across the cell to be secreted at the apical surface. During this process, a cleavage occurs that separates the extracellular (known as the secretory component) from the transmembrane segment , Through its N-linked glycans ensures anchoring of secretory IgA (sIgA) molecules to mucus lining the epithelial surface to neutralize extracellular pathogens (PubMed:12150896).
