Product Description: Phosphatase and actin regulator 3 (PHACTR3), also known as scapinin, is a multifunctional protein that binds both actin and the catalytic subunit of protein phosphatase 1 (PP1), inhibiting PP1 activity and thus serving as a regulatory subunit of this phosphatase. It is associated with the nuclear scaffold in proliferating cells but predominantly localizes to the plasma membrane in adherent cells, where it influences cell shape, motility, and spreading by regulating membrane-cytoskeleton dynamics. The N-terminal region of PHACTR3 contains conserved basic and hydrophobic residues critical for its membrane binding and interaction with acidic phospholipids, enabling its role in actin cytoskeleton remodeling. Alternative splicing produces multiple isoforms with potentially distinct functions. Dysregulation of PHACTR3 has been implicated in neuronal morphology alterations and may contribute to pathological conditions involving cytoskeletal defects and impaired cell motility. Its role in modulating PP1 activity and actin dynamics positions PHACTR3 as a key player in cellular processes relevant to cancer metastasis and neurodevelopmental disorders.
