Product Description: Neurotrophic receptor tyrosine kinase 3 (NTRK3), also known as TrkC, is a membrane-bound receptor tyrosine kinase that specifically binds neurotrophin-3 (NT-3) with high affinity. Structurally, NTRK3 consists of an extracellular ligand-binding domain, a single transmembrane helix, and an intracellular tyrosine kinase domain. Upon NT-3 binding, NTRK3 dimerizes and autophosphorylates, activating downstream signaling pathways such as PI3K/AKT, MAPK, and PLCgamma, which regulate neuronal survival, differentiation, proliferation, and synaptogenesis. Multiple isoforms of NTRK3 exist due to alternative splicing, with some lacking kinase activity but still influencing neuronal differentiation. NTRK3 also functions as a dependence receptor, promoting apoptosis in the absence of ligand. Clinically, NTRK3 is implicated in various cancers including medulloblastoma, secretory breast carcinoma, and upper tract urothelial carcinomas, often through gene fusions or altered expression. Its promoter methylation status serves as a biomarker in colorectal and esophageal cancers. Protein Function: Receptor tyrosine kinase involved in nervous system and probably heart development.
