Product Description: Neutrophil cytosolic factor 2 (NCF2) is a 67-kilodalton cytosolic protein encoded by the NCF2 gene located on chromosome 1 in humans. It functions as a critical subunit of the multi-protein NADPH oxidase complex found in neutrophils, mediating the transfer of electrons from cytosolic NADPH to molecular oxygen to produce superoxide anions. This process generates a respiratory burst that delivers superoxide to the lumen of neutrophil phagosomes, playing an essential role in the immune response against pathogens. Mutations in NCF2 can result in chronic granulomatous disease, characterized by impaired neutrophil function and recurrent infections. Additionally, certain genetic variations in NCF2 have been associated with increased risk of systemic lupus erythematosus, an autoimmune disorder. These variants are thought to impair NADPH oxidase function, reducing reactive oxygen species production and triggering an overactive immune response that may attack the body's own tissues and organs. Protein Function: Subunit of the phagocyte NADPH oxidase complex that mediates the transfer of electrons from cytosolic NADPH to O2 to produce the superoxide anion (O2(-)) (PubMed:12207919, PubMed:38355798). In the activated complex, electrons are first transferred from NADPH to flavin adenine dinucleotide (FAD) and subsequently transferred via two heme molecules to molecular oxygen, producing superoxide through an outer-sphere reaction (PubMed:38355798). Activation of the NADPH oxidase complex is initiated by the assembly of cytosolic subunits of the NADPH oxidase complex with the core NADPH oxidase complex to form a complex at the plasma membrane or phagosomal membrane (PubMed:38355798). This activation process is initiated by phosphorylation dependent binding of the cytosolic NCF1/p47-phox subunit to the C-terminus of CYBA/p22-phox (By similarity) [The Uniprot Consortium]
