Product Description: Myosin XVI (MYO16) is a vertebrate-specific, neuronally expressed unconventional myosin predominantly found in neural tissues, especially during late embryonic and early postnatal development when neuronal migration and dendritic elaboration occur. Structurally, MYO16 features an N-terminal ankyrin repeat domain that binds protein phosphatase 1, a myosin motor domain with likely impaired ATPase activity, and a tail domain that interacts with phosphoinositide 3-kinase (PI3K) and the WAVE regulatory complex (WRC), both crucial for actin cytoskeleton dynamics. MYO16 acts as a signal transduction element, integrating pathways that regulate actin reorganization at postsynaptic sites and may also play a role in cell cycle regulation, with two main isoforms: the full-length Myo16b (cytoplasmic and nuclear) and the tailless Myo16a (cytoplasmic). Dysregulation or impaired expression of MYO16 has been associated with neuropsychiatric disorders such as autism, schizophrenia, and bipolar disorder. Protein Function: Myosins are actin-based motor molecules with ATPase activity. Unconventional myosins serve in intracellular movements. Their highly divergent tails are presumed to bind to membranous compartments, which would be moved relative to actin filaments. May be involved in targeting of the catalytic subunit of protein phosphatase 1 during brain development. Activates PI3K and concomitantly recruits the WAVE1 complex to the close vicinity of PI3K and regulates neuronal morphogenesis (By similarity) [The Uniprot Consortium]
