Product Description: MAX dimerization protein 4 (MXD4), also known as MAD4, is a basic helix-loop-helix-leucine zipper (bHLH-Zip) transcriptional repressor that belongs to the MAD gene family. MXD4 heterodimerizes with the MAX protein to form a sequence-specific DNA-binding complex that recognizes the core sequence 5'-CAC[GA]TG-3', thereby repressing transcription. This repression antagonizes MYC transcriptional activity by competing for MAX binding, which is crucial because MYC-MAX complexes act as transcriptional activators. Through this mechanism, MXD4 plays a significant role in regulating cell growth and differentiation, and studies in rodents suggest that MAD family proteins, including MXD4, function as tumor suppressors. Gene Ontology annotations highlight its protein dimerization and transcription corepressor activities. Disease associations for MXD4 include colon sarcoma and bladder sarcoma, indicating that dysregulation of its function may contribute to tumorigenesis, likely due to its role in suppressing MYC-dependent cell transformation.
