Product Description: Leukocyte receptor tyrosine kinase (LTK) is a receptor-type protein tyrosine kinase of 864 amino acids, structurally composed of a 423-amino acid extracellular domain, a transmembrane domain, and a 415-amino acid cytoplasmic kinase domain. It belongs to the ros/insulin receptor kinase family and primarily functions in signal transduction pathways regulating cell growth, differentiation, and the secretory pathway, particularly involving endoplasmic reticulum export sites and ER-to-Golgi transport. LTK is activated by ligands ALKAL1 and ALKAL2, leading to phosphorylation and activation of downstream pathways like MAPK and PI3 kinase. It is expressed in hematopoietic cells, pre-B lymphocytes, neurons, and the adult hippocampus, influencing neuronal development and neurite outgrowth. Pathologically, LTK has been implicated in autoimmune diseases like systemic lupus erythematosus (SLE), glioma susceptibility, and is often upregulated in acute myeloid leukemia. Its signaling role in disease contexts relates to aberrant kinase activity and mutations affecting cell survival and proliferation. Protein Function: Receptor with a tyrosine-protein kinase activity (PubMed:10445845, PubMed:20548102, PubMed:30061385). Following activation by ALKAL1 or ALKAL2 ligands at the cell surface, transduces an extracellular signal into an intracellular response (PubMed:30061385, PubMed:34646012). Ligand-binding to the extracellular domain induces tyrosine kinase activation, leading to activation of the mitogen-activated protein kinase (MAPK) pathway (PubMed:20548102). Phosphorylates almost exclusively at the first tyrosine of the Y-x-x-x-Y-Y motif (By similarity). The exact function of this protein is not known, studies with chimeric proteins demonstrate its ability to promote growth and specifically neurite outgrowth, and cell survival (PubMed:18849880, PubMed:9223670).
