Product Description: LIM and calponin homology domains 1 (LIMCH1) is an actin stress fiber-associated protein characterized by an N-terminal calponin homology (CH) domain, a C-terminal LIM domain, and central coiled-coil regions. The CH domain facilitates actin binding, while the LIM domain mediates protein interactions. LIMCH1 regulates the activity of non-muscle myosin IIa (NM-IIA) by promoting phosphorylation of myosin regulatory light chain (MRLC), enhancing actomyosin contractility and stress fiber assembly. This function stabilizes focal adhesions and modulates cell morphology and motility. LIMCH1 depletion reduces MRLC phosphorylation, disrupts stress fibers, and decreases focal adhesion stability, leading to increased cell spreading and migration, indicating its role in restricting cell motility. In disease contexts, alterations in LIMCH1 expression may influence cancer cell migration and metastasis. Protein Function: Actin stress fibers-associated protein that activates non-muscle myosin IIa. Activates the non-muscle myosin IIa complex by promoting the phosphorylation of its regulatory subunit MRLC/MYL9. Through the activation of non-muscle myosin IIa, positively regulates actin stress fibers assembly and stabilizes focal adhesions.
