Product Description: Linker for Activation of T Cells (LAT) is a transmembrane adaptor protein crucial for transducing intracellular signals from the TCR/CD3 complex in T cells. LAT has a short extracellular N-terminal fragment, a hydrophobic transmembrane segment, and a long cytoplasmic tail containing nine conserved tyrosine residues. Upon TCR engagement, LAT becomes rapidly tyrosine-phosphorylated, creating binding sites for various signaling proteins such as PLC-gamma1, Grb2, Gads, and SLP-76, which are essential for downstream signaling cascades. LAT is localized to the glycolipid-enriched membrane (GEM) subdomains of the plasma membrane by palmitoylation. Defects in LAT function lead to impaired T cell activation, including reduced intracellular calcium, Ras activation, and IL-2 production. LAT is also involved in regulating T cell memory differentiation and is necessary for memory CD8 T cells to proliferate and produce cytokines upon antigen re-exposure. Protein Function: Required for TCR (T-cell antigen receptor)- and pre-TCR-mediated signaling, both in mature T-cells and during their development (PubMed:23514740, PubMed:25907557).
