Product Description: IL-16 is encoded by the IL16 gene located on chromosome 15q26.3. It is initially produced as a precursor protein (pro-IL-16) that is cleaved by caspase-3 to generate the mature, secreted form of IL-16. Both pro-IL-16 and mature IL-16 are biologically active. IL-16 functions as a chemoattractant for CD4+ T cells, monocytes, and eosinophils, and can also regulate cell growth and differentiation. Increased levels of IL-16 have been associated with various inflammatory and autoimmune diseases, including multiple sclerosis, rheumatoid arthritis, and antineutrophil cytoplasmic antibody-associated vasculitis. Genetic polymorphisms in the IL16 gene, such as rs11556218, have also been linked to an increased risk of certain cancers, as well as other conditions like diabetes and osteoarthritis. Overall, IL-16 appears to play a complex role in modulating immune and inflammatory responses, with implications for both disease pathogenesis and potential therapeutic targeting. Protein Function: Interleukin-16 stimulates a migratory response in CD4+ lymphocytes, monocytes, and eosinophils. Primes CD4+ T-cells for IL-2 and IL-15 responsiveness. Also induces T-lymphocyte expression of interleukin 2 receptor. Ligand for CD4 , May act as a scaffolding protein that anchors ion channels in the membrane , Isoform 3 is involved in cell cycle progression in T-cells. Appears to be involved in transcriptional regulation of SKP2 and is probably part of a transcriptional repression complex on the core promoter of the SKP2 gene. May act as a scaffold for GABPB1 (the DNA-binding subunit the GABP transcription factor complex) and HDAC3 thus maintaining transcriptional repression and blocking cell cycle progression in resting T-cells [The Uniprot Consortium]
