Product Description: GIT ArfGAP 1 (GIT1), encoded by the GIT1 gene, is a multidomain scaffolding protein that regulates intracellular signaling, vesicular trafficking, and cytoskeletal dynamics. It functions as an Arf GTPase-activating protein (ArfGAP), catalyzing GTP hydrolysis in ADP-ribosylation factor (ARF) proteins to modulate membrane trafficking and receptor internalization. GIT1 contains an N-terminal ArfGAP domain, ankyrin repeats, a Spa2 homology domain (SHD), and a coiled-coil region, enabling interactions with partners like PAK kinases, PIX proteins, and focal adhesion complexes. These interactions position GIT1 as a critical node in signaling pathways controlling cell adhesion, migration, and synaptic plasticity. GIT1 also localizes to postsynaptic densities, influencing dendritic spine morphogenesis and neurotransmission. Dysregulation of GIT1 is linked to neuropsychiatric disorders, particularly attention deficit hyperactivity disorder (ADHD) and schizophrenia, with studies implicating altered GIT1 expression or function in synaptic deficits underlying these conditions. Additionally, GIT1 interacts with beta-arrestin in GPCR signaling, further connecting it to neuronal and developmental processes.
