Product Description: Fas-associated factor 1 (FAF1) is a 74-kDa multifunctional scaffolding protein involved in apoptosis and cellular regulation. It binds directly to the Fas receptor and components of the Fas-death-inducing signaling complex (Fas-DISC) such as FADD and caspase-8, facilitating Fas-mediated apoptosis by acting upstream of caspase-8. FAF1 contains multiple ubiquitin-related domains (UBA, UBL1, UBL2, UBX) which enable interactions with ubiquitinated proteins and the valosin-containing protein complex, implicating it in ubiquitin-mediated proteasomal degradation and endoplasmic reticulum-associated protein degradation (ERAD). FAF1 also negatively regulates the NF-kappaB signaling pathway by inhibiting IKK activation and preventing nuclear translocation of NF-kappaB. Dysregulation of FAF1 is linked to diseases including cancer, where decreased FAF1 expression may contribute to tumor progression, and neurodegenerative disorders through its role in cell survival and death. FAF1 additionally influences antiviral immune responses by regulating type I interferon production during RNA virus infections, highlighting its diverse roles in cell death and immune signaling. Required for the progression of DNA replication forks by targeting DNA replication licensing factor CDT1 for degradation (PubMed:26842564).
