Product Description: Deoxyhypusine hydroxylase (DOHH) is a crucial enzyme involved in the post-translational modification of the eukaryotic translation initiation factor 5A (eIF5A), specifically catalyzing the hydroxylation of deoxyhypusine to form hypusine. This modification is essential for eIF5A's role in protein synthesis and cell proliferation. DOHH is a nonheme diiron monooxygenase, characterized by its ability to activate molecular oxygen for substrate hydroxylation, and features a stable peroxo-diiron(III) intermediate that has been crystallized for structural studies. Mutations in the DOHH gene have been linked to neurodevelopmental disorders, highlighting its importance in cellular processes and development. The enzyme's activity is also considered a potential therapeutic target for various diseases, including cancer and metabolic disorders, due to its role in regulating eIF5A function and cellular growth pathways. Protein Function: Catalyzes the hydroxylation of the N(6)-(4-aminobutyl)-L-lysine intermediate produced by deoxyhypusine synthase/DHPS on a critical lysine of the eukaryotic translation initiation factor 5A/eIF-5A. This is the second step of the post-translational modification of that lysine into an unusual amino acid residue named hypusine (PubMed:16371467, PubMed:16533814, PubMed:19706422).
