Product Description: Carboxypeptidase A2 (CPA2) is a zinc-dependent metalloprotease encoded by the CPA2 gene (chromosome 7q32.2) and primarily synthesized in the pancreas as a proenzyme (zymogen). Secreted into the duodenum, it is activated by trypsin cleavage of its N-terminal propeptide, generating a mature enzyme (~45 kDa) that hydrolyzes C-terminal hydrophobic amino acids from dietary proteins, aiding digestion. Structurally, CPA2 features a catalytic domain with a conserved zinc-binding motif (His-Glu-X-X-His) and a substrate-binding pocket that dictates its specificity. Unlike CPA1, CPA2 exhibits broader substrate tolerance, including aromatic and aliphatic residues. Elevated serum CPA2 levels are associated with pancreatic disorders like acute pancreatitis and pancreatic ductal adenocarcinoma. Rare CPA2 variants are implicated in hereditary pancreatitis. CPA2 also acts as a minor food allergen, linked to wheat-dependent exercise-induced anaphylaxis.
