Product Description: Basal cell adhesion molecule (BCAM), also known as Lutheran blood group glycoprotein or CD239, is a transmembrane glycoprotein of the immunoglobulin superfamily comprising five extracellular Ig-like domains, a single transmembrane domain, and a short cytoplasmic tail. It functions primarily as a receptor for laminin alpha5, an extracellular matrix protein, mediating cell adhesion, motility, migration, and invasion by interacting with laminin and cytoskeletal proteins such as spectrin. BCAM is widely expressed in red blood cells, endothelial, smooth muscle, and epithelial cells and plays a critical role in cell signaling and cytoskeletal organization. Dysregulation or abnormal expression of BCAM is implicated in several diseases, notably sickle cell anemia, where it contributes to vaso-occlusion, and various cancers including liver, lung, colorectal, bladder, and ovarian cancers, where it promotes tumor cell migration and metastasis. Protein Function: Transmembrane glycoprotein that functions as both a receptor and an adhesion molecule playing a crucial role in cell adhesion, motility, migration and invasion (PubMed:9616226, PubMed:31413112). Extracellular domain enables binding to extracellular matrix proteins, such as laminin, integrin and other ligands while its intracellular domain interacts with cytoskeletal proteins like hemoglobin, facilitating cell signal transduction (PubMed:17158232). Serves as a receptor for laminin alpha-5/LAMA5 to promote cell adhesion (PubMed:15975931). Mechanistically, JAK2 induces BCAM phosphorylation and activates its adhesion to laminin by stimulating a Rap1/AKT signaling pathway in the absence of EPOR (PubMed:23160466) [The Uniprot Consortium]
