Product Description: Caspase recruitment domain family member 14 (CARD14) is a scaffold protein belonging to the membrane-associated guanylate kinase (MAGUK) family and the CARD-CC protein family, characterized by a caspase-associated recruitment domain (CARD) and a coiled-coil (CC) domain. CARD14 is primarily expressed in the skin, where it plays a crucial role in assembling multiprotein complexes at specialized plasma membrane regions and activating the NF-kappaB signaling pathway, which regulates genes involved in immune responses, inflammation, and cell survival. The protein interacts with BCL10 and MALT1 to form a CBM signalosome, facilitating NF-kappaB activation and modulating skin cell homeostasis, including the regulation of filaggrin (FLG) expression. Pathogenic mutations in CARD14 are associated with several skin disorders, most notably psoriasis and pityriasis rubra pilaris, as well as atopic eczema, due to dysregulated inflammatory signaling in the skin. Protein Function: Acts as a scaffolding protein that can activate the inflammatory transcription factor NF-kappa-B and p38/JNK MAP kinase signaling pathways. Forms a signaling complex with BCL10 and MALT1, and activates MALT1 proteolytic activity and inflammatory gene expression. MALT1 is indispensable for CARD14-induced activation of NF-kappa-B and p38/JNK MAP kinases (PubMed:11278692, PubMed:21302310, PubMed:27071417, PubMed:27113748). May play a role in signaling mediated by TRAF2, TRAF3 and TRAF6 and protects cells against apoptosis , Not able to activate the inflammatory transcription factor NF-kappa-B and may function as a dominant negative regulator (PubMed:21302310, PubMed:26358359) [The Uniprot Consortium]
