Product Description: Cell Adhesion Molecule 3 (CADM3) is a protein encoded by the CADM3 gene, which plays a crucial role in cell-cell adhesion, particularly in neural tissues. It is a member of the CADM family and is expressed in axons, where it interacts primarily with CADM4 to facilitate axon guidance, myelination, and maintenance of axonal architecture. CADM3 exhibits calcium-independent homophilic and heterophilic cell-cell adhesion activities, and its interaction with other proteins like DLG3, PALS2, and CASK suggests its involvement in membrane-associated guanylate kinase (MAGUK) subfamily member-binding activity. Mutations in the CADM3 gene have been associated with Charcot-Marie-Tooth disease type 2FF (CMT2FF), a neurodegenerative disorder characterized by axonal degeneration and demyelination. Specifically, a Y172C substitution in the extracellular domain of CADM3 has been identified as a causative mutation for CMT2FF, leading to structural protein rearrangement and impaired cell adhesion. Protein Function: Involved in cell-cell adhesion. Has both calcium-independent homophilic cell-cell adhesion activity and calcium-independent heterophilic cell-cell adhesion activity with IGSF4, NECTIN1 and NECTIN3.
