Product Description: BRISC and BRCA1 A complex member 1 (BABAM1), also known as MERIT40 or NBA1, is a protein encoded by the BABAM1 gene that serves as a critical component of both the BRCA1-A and BRISC multiprotein complexes. BABAM1 is essential for maintaining the integrity and localization of the BRCA1-A complex at sites of DNA double-strand breaks (DSBs), where it specifically recognizes Lys-63-linked ubiquitinated histones H2A and H2AX. This complex exhibits deubiquitinase activity, removing Lys-63-linked ubiquitin chains, which helps target the BRCA1-BARD1 heterodimer to DNA damage sites and regulates DNA repair pathway choice by promoting non-homologous end joining over homologous recombination. In the BRISC complex, BABAM1 stabilizes BABAM2 and supports Lys-63-linked deubiquitinase activity mediated by BRCC3/BRCC36, contributing to normal mitotic spindle assembly and microtubule-kinetochore attachment. Additionally, BABAM1 modulates interferon signaling by deubiquitinating the interferon receptor IFNAR1, enhancing its stability and downregulating bacterial lipopolysaccharide responses. Dysfunction or altered expression of BABAM1 impacts DNA repair fidelity and cellular stress responses, implicating it in cancer susceptibility and genomic instability.
