Product information "Anti-Human ATAT1 Antibody Pair"
Product Description: Alpha tubulin acetyltransferase 1 (ATAT1) is a key enzyme responsible for the acetylation of alpha-tubulin at lysine 40, a post-translational modification that stabilizes microtubules and enhances their resistance to mechanical stress. Structurally, ATAT1 shares similarities with Gcn5 histone acetyltransferases but features a unique substrate-binding groove that confers specificity for alpha-tubulin. It plays crucial roles in cellular processes such as cytoskeletal organization, intracellular trafficking, mitosis, and cell motility. ATAT1 activity is tightly regulated, and mutations affecting its catalytic residues significantly alter tubulin acetylation levels in vivo. Dysregulation of ATAT1-mediated tubulin acetylation is implicated in various diseases, including neurological disorders like Charcot-Marie-Tooth disease, Parkinson's, and Alzheimer's disease, as well as cancers such as breast, pancreatic, and head and neck squamous cell carcinoma. In the nervous system, ATAT1 influences axonal growth and regeneration, with reduced ATAT1 linked to impaired neurite outgrowth and axon injury. Protein Function: Specifically acetylates 'Lys-40' in alpha-tubulin on the lumenal side of microtubules. Promotes microtubule destabilization and accelerates microtubule dynamics, this activity may be independent of acetylation activity. Acetylates alpha-tubulin with a slow enzymatic rate, due to a catalytic site that is not optimized for acetyl transfer. Enters the microtubule through each end and diffuses quickly throughout the lumen of microtubules. Acetylates only long/old microtubules because of its slow acetylation rate since it does not have time to act on dynamically unstable microtubules before the enzyme is released. Required for normal sperm flagellar function. Promotes directional cell locomotion and chemotaxis, through AP2A2-dependent acetylation of alpha-tubulin at clathrin-coated pits that are concentrated at the leading edge of migrating cells. May facilitate primary cilium assembly [The Uniprot Consortium]
