CREB is a b-ZIP transcription factor that activates target genes through cAMP response elements. CREB is able to mediate signals from numerous physiological stimuli, resulting in regulation of a broad array of cellular responses. While CREB is expressed in numerous tissues, it plays a large regulatory role in the nervous system. CREB is believed to play a key role in promoting neuronal survival, precursor proliferation, neurite outgrowth and neuronal differentiation in certain neuronal populations. CREB promotes outgrowth and differentiation as a mediator of neurotrophin pathways. CREB is able to selectively activate numerous downstream genes through interactions with different dimerization partners. CREB is activated by phosphorylation at Ser133 by various signaling pathways including Erk, Ca2+ and stress signaling. Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry and Chromatin Immunopreciptation. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000 Detected phosphorylated CREB in RIPA lysates from mouse 3T3 cells treated with 50mM forskolin. PC-12 and forskolin treated PC-12 cell lysates were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho-CREB (1:1000). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. , Immunoprecipitation: 10ul immunoprecipitates phosphorylated CREB from 500ug of rat PC-12 cells treated with 50uM forskolin, Immunohistochemistry: 1:1000 dilution detects phosphorylated CREB in formalin-fixed paraffin-embedded normal and ischemic rat brain sections., Chromatin Immunopreciptation: A representative lot immunoprecipitated CREB using chromatin from 293T cells., Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
