Anti-Osteoblast Specific Factor 2 (OSF-2, Periostin, Fasciclin-I like)

Osteoblast Specific Factor 2 (OSF-2), also know as Periostin, is a secreted heparin-binding glycoprotein originally identified as an osteoblast-specific factor. It has since been found to be expressed in embryonic heart. OSF-2 is highly homologous to TGF-Beta1-inducible protein Beta-Ig-H3 and both proteins contain four fasciclin domains. OSF-2 mediates cell adhesion by binding to integrins and serves as a useful marker of mesenchymal cells in the developing heart. Mouse and human OSF-2 share 89% amino acid sequence identity. Applications: Suitable for use in ELISA, Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1-2ug/ml. Using a colorimetric detection system, the detection limit is ~25ng/ lane (non-reducing conditions) and 50ng/lane (reducing conditions). Chemiluminescent detection will increase sensitivity ~5-50 fold., Direct ELISA: 0.5-1ug/ml Detection limit for rmOSF-2 is ~3ng/well., Immunocytochemistry: 10ug/ml detects OSF2 in mouse ST2 cells differentiated to osteocytes. Cells were fixed with PBS containing 4% paraformaldehyde for 20 minutes at RT and blocked with PBS, 10% normal donkey serum, 0.1% Triton X-100, 1% BSA for 45 minutes at RT. After blocking, cells were incubated with diluted O8057-04 overnight at 4°C followed by IgG (FITC) anti-rat at RT in the dark for 1 hour. Between each step, cells were washed with PBS, 0.1% BSA., Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
