NLN (neurolysin) is a metalloproteinase of the MA(E) clade, in the M3 family. The name is derived from the ability to degrade neurotensin. The M3 family includes the structurally similar thimet oligopeptidase (EC 3.4.24.15), and contains the canonical HExxHxxH zinc binding motif in the catalytic domain, but with the third histidine three residues closer in than the MMP family. Neurolysin has been reported in the cytosol and in mitochondria, and in pig, rat and chimp there are alternatively spliced versions that target neurolysin to the mitochondria. Applications: Suitable for use in ELISA, Immunoprecipitation, and Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml, ELISA: 0.5-1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
