This monoclonal antibody recognizes the 56.5kD (CK10), 50kD (CK14), 50kD (CK15), 48kD (CK16), 40kD (CK19) keratins of the acidic (Type I or LMW) subfamily. Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI <5.7) and basic (pI >6.0) subfamilies. The acidic keratins have molecular weights (MW) of 56.5, 55, 51, 50, 50', 48, 46, 45, and 40kD. Monoclonal antibody AE3 recognizes the 65-67, 64, 59, 58, 56, and 52kD keratins of basic subfamily. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. AE1/AE3 is a broad spectrum anti pan-keratin antibody cocktail, which differentiates epithelial tumors from non-epithelial tumors e.g. squamous vs. adenocarcinoma of the lung, liver carcinoma, breast cancer, and esophageal cancer. Applications: Suitable for use in ELISA, Flow Cytometry, Western Blot, Immunofluorescence and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5-1ug/ml, Flow Cytometry: 0.5-1ug/10e6 cells, Immunofluorescence: 1-2ug/ml, Immunohistochemistry (Frozen & Formalin fixed paraffin embedded): 0.5-1ug/ml/30min @ RT, Optimal dilutions to be determined by the researcher. Positive Control: Skin, Squamous cell carcinoma (SCC), Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
