CD97 gene is a member of the EGF-TM7 family of class II seven-span transmembrane (7-TM) molecules, likely encoded by a gene cluster on the short arm of chromosome 19. The encoded product is a glycoprotein that is present on the surface of most activated leukocytes and spans the membrane seven times, which is a defining feature of G protein-coupled receptors. The protein has an extended extracellular region with several N-terminal epidermal growth factor (EGF)-like domains, which mediate binding to its cellular ligand, decay accelerating factor (DAF, CD55), a regulatory protein of the complement cascade. The presence of structural features characteristic of extracellular matrix proteins and transmembrane proteins suggests that this protein is a receptor involved in both cell adhesion and signaling processes early after leukocyte activation. Alternative splicing has been observed for this gene and three variants have been found. CD97 antigen is a receptor involved in both adhesion and signaling processes early after leukocyte activation. It also plays an essential role in leukocyte migration. The unprocessed precursor is 853 amino acids long and has a predicted molecular weight of 91.9KDa. CD97 antigen is a heterodimer consisting of a large extracellular alpha subunit and a seven-transmembrane beta subunit that are non-covalently linked. It contains five EGF-like domains and one GPS domain. This protein interacts with complement decay-accelerating factor (DAF) through the first two EGF domains, and with chondroitin sulfate (only isoform 1) through the fourth EGF domain. CD97 antigen is broadly expressed and it is present on most hematopoietic cells. It is also abundantly expressed by smooth muscle cells. Expression of this protein is increased under inflammatory conditions in the CNS of multiple sclerosis and in synovial tissue of patients of rheumatoid arthritis. This protein is rapidly up-regulated during lymphocyte activation. C97 antigen belongs to the G-protein coupled receptor 2 fmaily and the LN-TM7 subfamily. , Applications: , Suitable for use in Western Blot and ELISA. Other applications have not been tested. Recommended Dilutions: ELISA: 0.1ng/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile 40-50% glycerol, ddH2O. Reconstituted product is stable for 12 months at -20°C. Aliquot and store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
